제목: Retinal regeneration - a new therapeutic paradigm for retinal degenerative diseases
초록: Retinal neurons are generated during development and degenerate during lifetime. Degenerated cells can be replaced by new cells in many animal tissues that contain stem cells. Given the absence of residual retinal stem cells, degenerated retinal neurons cannot be regenerated in the retina, causing various retinal degenerative diseases. However, retinal neurons can be regenerated from Müller glia (MG) in cold-blooded vertebrates upon the injury, highlight the incompetency of mammalian MG for retinal degeneration. We found that prospero-related homeobox (Prox1), a well-known suppressor of neural stem cell, accumulated in MG and suppresses injury-induced MG reprogramming to retinal progenitor cells in mouse retinas. Consistent with this, we succeeded in inducing reprogramming of MG in the mouse retina by blocking the accumulation of Prox1, consequently delaying vision loss in the retinitis pigmetosa disease model mice. Collectively, our study suggests that MG reprogramming and subsequent retinal neurogenesis in the injured mammalian retina can be recovered by eliminating PROX1 in MG by various therapeutic strategies.
